目的探讨三氧化二砷联合氟尿嘧啶(5-FU)对肝癌细胞凋亡相关因子Bcl-2和Bax表达的影响。方法体外培养人肝癌SMMC-7721细胞株,将细胞分为四组,即As2O3组、5-FU组、As2O3+5-FU组及对照组,采用免疫细胞化学法检测各组SMMC-7721细胞Bcl-2、Bax蛋白的表达。结果 As2O3组、5-FU组、As2O3+5-FU组细胞Bcl-2蛋白表达较对照组明显降低,Bax蛋白表达明显升高(P<0.01);联合用药组细胞Bcl-2蛋白表达较单用药组明显降低,Bax蛋白表达明显升高(P<0.01)。结论 As2O3联合5-FU可下调Bcl-2蛋白的表达,上调Bax蛋白的表达,从而诱导肝癌细胞凋亡。
Objective To investigate the effects of arsenic trioxide combined with 5-fluorouracil on the expression of apoptosis-related factors Bcl-2 and Bax in hepatocellular carcinoma. Methods Human hepatoma SMMC-7721 cells were cultured in vitro and divided into four groups: As2O3 group, 5-FU group, As2O3 + 5-FU group and control group. Immunocytochemistry was used to detect the expression of Bcl- -2, Bax protein expression. Results The Bcl-2 protein expression in As2O3 group, 5-FU group and As2O3 + 5-FU group was significantly lower than that in control group and Bax protein expression (P <0.01) The treatment group was significantly lower, Bax protein expression was significantly increased (P <0.01). Conclusion As2O3 combined with 5-FU can down-regulate the expression of Bcl-2 protein and up-regulate the expression of Bax protein, thereby inducing hepatocellular carcinoma cell apoptosis.
