目的探讨糖基化终产物(AGEs)对体外培养兔视网膜Müller细胞的影响。方法培养兔视网膜Müller细胞,体外制备AGEs及其对照物,实验分为牛血清白蛋白AGE(AGE-BSA)作用3 d组、AGE-BSA作用9 d组、AGE-BSA对照3 d组、AGE-BSA对照9 d组、空白对照3 d组及空白对照9 d组。采用光镜下观察及流式细胞仪检测AGEs对Müller细胞凋亡发生率的影响。结果AGEs对Müller细胞作用3 d后,与对照组相比,细胞形态和数量无明显变化,无明显细胞凋亡;而作用9 d后,细胞数量明显减少,细胞凋亡率明显增高。结论AGEs可抑制视网膜Müller细胞的生长。
Objective To investigate the effect of advanced glycation end products (AGEs) on cultured rabbit retinal Müller cells. METHODS: Rabbit retinal Müller cells were cultured and AGEs and their controls were prepared in vitro. The experiment was divided into three groups: AGE-BSA (AGE-BSA) for 3 d, AGE-BSA for 9 d, AGE-BSA for 3 d, AGE -BSA control 9 d group, blank control 3 d group and blank control 9 d group. The effects of AGEs on the apoptosis of Müller cells were detected by light microscopy and flow cytometry. Results After treated with AGEs for 3 days, the cell morphology and number of Müller cells did not change significantly compared with that of the control group, and no significant apoptosis was observed. However, after 9 days, the number of cells was significantly decreased and the apoptosis rate was significantly increased. Conclusion AGEs can inhibit the growth of retinal Müller cells.
