目的研究不同发育阶段的豚鼠睾丸和附睾组织中激素敏感性脂肪酶(HSL)的表达及活性与生殖能力成熟的关系。方法取不同年龄阶段的豚鼠(6d的幼鼠、16d青春期豚鼠以及70d的成年豚鼠)用RT-PCR方法检测睾丸和附睾组织中HSL的mRNA表达,37℃下比色法对酶活性进行测定,放免法测定血清中睾酮水平,HE染色睾丸和附睾组织切片进行组织形态学分析。结果发育阶段的豚鼠随着天龄的增加,睾丸特异性HSLmRNA表达也增加(P<0.05),酶活性增强(P<0.05),血清中睾酮水平升高(P<0.01),组织学显示16d龄的豚鼠睾丸和附睾中开始出现精子,70d龄精子发育成熟。HSL的表达和酶活性与血清睾酮水平高度相关(r=0.75,P<0.01;r=0.96,P<0.01)。结论发育成熟豚鼠睾丸和附睾组织有较强的HSL表达及活性,HSL与睾丸的发育成熟以及精子发生可能存在相关关系。
Objective To study the expression and activity of hormone-sensitive lipase (HSL) in the testis and epididymis of guinea pigs at different developmental stages in relation to reproductive maturity. Methods The mRNA expression of HSL in testis and epididymal tissue was detected by RT-PCR in guinea pigs (6d, 16d, and 70d) of adult guinea pigs of different ages. The enzyme activity was determined by colorimetry at 37 ℃, Serum testosterone levels were determined by radioimmunoassay, HE staining of testicular and epididymal sections for histomorphological analysis. Results The testis-specific HSLmRNA expression was also increased (P <0.05), the enzyme activity was enhanced (P <0.05) and the level of testosterone in serum was increased (P <0.01) Sperms began to appear in the testis and epididymis of guinea pigs at the age of 60 days, and spermatozoa matured at 70 days. HSL expression and enzyme activity were highly correlated with serum testosterone levels (r = 0.75, P <0.01; r = 0.96, P <0.01). Conclusion There is a strong HSL expression and activity in mature testis and epididymis of mature guinea pigs. There may be correlation between HSL and testicular maturation and spermatogenesis.
