目的:检测新生儿脐血及成人外周血中CD4+ CD25high Foxp3+调节性T细胞(Treg)的比例,探讨Treg在新生儿脐血中的临床意义。方法:采用流式细胞术(FCM)检测30例新生儿脐血单个核细胞中CD4+T细胞、CD4+ CD25+/CD4+、CD4+ CD25high/CD4+T细胞百分比以及绝对计数,进一步分析CD4+ CD25+及CD4+ CD25highT细胞中Foxp3+的百分比,以及利用RT-PCR方法检测Foxp3 mRNA表达水平,并与27例成人外周血单个核细胞(PBMC)中的上述指标进行比较。结果:与成人PBMC比较,新生儿脐血单个核细胞中CD4+T细胞百分比及绝对计数均增高(P<0.01);CD4+ CD25+/CD4+及CD4+ CD25high/CD4+百分比减低(P<0.05),但绝对计数增高(P<0.01);新生儿脐血CD4+ CD25+及CD4+ CD25high T细胞中Foxp3+表达较成人外周血减低(P<0.01);新生儿脐血Foxp3 mRNA表达水平亦较成人外周血减低(P<0.05)。结论:新生儿脐血中存在单纯且数量较多的CD4+ CD25high调节性T细胞,但Foxp3的表达水平低于成人外周血,它们可能具有独特的免疫调节作用,但功能尚未成熟。
Objective: To detect the proportion of CD4 + CD25high Foxp3 + regulatory T cells (Treg) in neonatal umbilical cord blood and adult peripheral blood and to explore the clinical significance of Treg in neonatal umbilical cord blood. Methods: The percentage of CD4 + T cells, CD4 + CD25 + / CD4 +, CD4 + CD25high / CD4 + T cells in cord blood mononuclear cells from 30 newborns were determined by flow cytometry (FCM), and the absolute counts were also analyzed. CD4 + CD25 + and CD4 + CD25highT The percentage of Foxp3 + in cells and the expression of Foxp3 mRNA by RT-PCR were compared with those in 27 peripheral blood mononuclear cells (PBMCs). Results: Compared with adult PBMC, the percentage and absolute count of CD4 + T cells in neonatal cord blood mononuclear cells increased (P <0.01), the percentage of CD4 + CD25 + / CD4 + and CD4 + CD25high / CD4 + (P <0.01). The expression of Foxp3 + in neonatal cord blood CD4 + CD25 + and CD4 + CD25high T cells was lower than that in adult peripheral blood (P <0.01), and the expression of Foxp3 mRNA in neonatal cord blood was also lower than that in adult peripheral blood (P < 0.05). CONCLUSION: There is a large number of CD4 + CD25high regulatory T cells in umbilical cord blood, but the expression level of Foxp3 is lower than that of adult peripheral blood. They may have unique immunomodulatory effects but their functions are not yet mature.
