目的:通过动物实验探讨平肝潜阳方治疗高血压病的中药药理学机制。方法:将40只原发性高血压大鼠(SHR)随机分为5组:模型组,平肝潜阳方高、中、低剂量组及马来酸依那普利对照组,每组8只;观察平肝潜阳方对SHR宏观表征和行为学的影响,测量给药前及给药后1、2、3、4周大鼠血压和心率,于给药4周后,检测大鼠血清一氧化氮(NO)和内皮素(ET)含量。结果:给药4周后,平肝潜阳方和马来酸依那普利对SHR宏观表征和行为学指标的影响表现出不同的特点,平肝潜阳方在改善大鼠急躁易怒、眩晕、头痛等方面具有一定的量效关系;与模型组相比,平肝潜阳方高、中剂量组能显著降低模型动物收缩压(SAP)、舒张压(DAP)和平均动脉压(MAP),减慢心率(P<0.01),2种药物均能显著增加大鼠血清NO含量,降低大鼠血清ET含量(P<0.01),但两者无显著差异。结论:平肝潜阳方不仅能够明显降低血压,而且能够改善大鼠的宏观表征,可能是通过调节血管活性物质而实现的。
OBJECTIVE: To explore the pharmacological mechanism of Pinggan Qianyang in the treatment of hypertension by animal experiments. Both drugs can significantly increase the content of NO in serum and decrease the content of ET in serum of rats (P <0.01), but there is no significant difference between them. Conclusion: Pinggan Qianyang can not only significantly lower blood pressure, but also improve macroscopic characterization of rats, possibly through regulating vasoactive substances.
