目的:探讨南瓜多糖对2型糖尿病小鼠血糖、血脂、血清超氧化物歧化酶、丙二醛及肝糖原的影响。方法:实验采用BALB/c小鼠制备2型糖尿病小鼠模型,成模小鼠随机分为模型组、药物对照组(二甲双胍,300mg/kg)、南瓜多糖组(150、300、600mg/kg),取10只健康小鼠为正常组。每日灌胃1次,28天后取血测定空腹血糖(FBG)、血脂(TG、TC、LDL-C、HDL-C)、血清超氧化物歧化酶(SOD)、丙二醛(MDA)及肝糖原含量,并行口服葡萄糖耐量试验(OGTT)。结果:南瓜多糖对正常小鼠血糖无影响,与糖尿病模型组小鼠比较,南瓜多糖3个剂量组可不同程度地降低糖尿病小鼠的FBG,改善糖耐量,提高肝糖原含量、SOD及HDL-C水平,降低TG、TC、LDL-C及MDA水平,差异具有统计学意义(P<0.01,P<0.05)。结论:南瓜多糖对正常小鼠无降糖作用,对2型糖尿病小鼠糖脂代谢紊乱具有改善作用,推测其机制与提高肝糖原含量及降低氧化应激水平有关。
Objective: To investigate the effects of pumpkin polysaccharide on blood glucose, blood lipid, serum superoxide dismutase, malondialdehyde and glycogen in type 2 diabetic mice. Methods: Type 2 diabetic mice were prepared by BALB / c mice. The model mice were randomly divided into model group, metformin group (300mg / kg), pumpkin polysaccharide group (150,300,600mg / kg) , Take 10 healthy mice as normal group. The blood samples were taken for 1 time daily and blood samples were taken for determination of fasting blood glucose (FBG), serum lipids (TG, TC, LDL-C, HDL-C), serum superoxide dismutase (SOD), malondialdehyde (MDA) Hepatic glycogen content, parallel oral glucose tolerance test (OGTT). Results: Compared with normal mice, the pumpkin polysaccharide had no effect on the blood glucose of diabetic mice. Compared with diabetic mice, the three doses of pumpkin polysaccharide could reduce the FBG, improve the glucose tolerance, improve the content of hepatic glycogen, SOD and HDL in diabetic mice (P <0.01, P <0.05). The levels of TG, TC, LDL-C and MDA were significantly lower than those of control group. Conclusion: Pumpkin polysaccharide has no hypoglycemic effect on normal mice and glucose-lipid metabolism disturbance in type 2 diabetic mice. It is speculated that the mechanism is related to increasing hepatic glycogen content and reducing oxidative stress.
