目的观察茶多酚对甲醛致人脐静脉内皮细胞氧化损伤的影响。方法体外常规培养内皮细胞,实验设正常对照组、甲醛损伤组(0.1 mmol/L)、茶多酚组(2.5、5、10、20 mg/L)。其中茶多酚先与细胞预孵12 h后加入甲醛,置CO2培养箱培养4 h,检测细胞培养液中超氧化物歧化酶(SOD)活力及乳酸脱氢酶(LDH)、丙二醛(MDA)、一氧化氮和细胞中谷胱甘肽含量,免疫细胞化学法检测核转录因子(NF-κB)。结果 2.5、5、10、20 mg/L茶多酚组LDH、MDA含量分别为2 021.02、1 878.17、1 748.56、1 625.72 U/L和3.50、3.30、2.99、2.68 nmol/mL,均分别低于甲醛损伤组,差异有统计学意义(P<0.01);SOD与GSH活力分别为24.16、24.67、25.56、26.02 U/mL和0.334、0.364、0.445、0.449μmol/mg prot均高于甲醛损伤组(P<0.01);茶多酚各组NF-κB活力与甲醛组比较明显下降(P<0.01)。结论茶多酚可提高内皮细胞抗氧化酶活性,减轻甲醛所致氧化损伤。
Objective To investigate the effect of tea polyphenols on the oxidative damage of human umbilical vein endothelial cells induced by formaldehyde. Methods The endothelial cells were cultured in vitro. The normal control group, formaldehyde injury group (0.1 mmol / L), and tea polyphenols group (2.5, 5, 10, 20 mg / L) were established. For the first time, tea polyphenols was added to the cells for 12 h, then the cells were incubated for 4 h in a CO2 incubator. The activities of superoxide dismutase (SOD), lactate dehydrogenase (LDH), malondialdehyde (MDA) ), Nitric oxide (NO) and glutathione (GSK) in cells, and nuclear factor-kappa B (NF-κB) by immunocytochemistry. Results The contents of LDH and MDA in 2.5, 5, 10, 20 mg / L tea polyphenols were 2021.02, 1878.17, 1748.56, 1625.72 U / L and 3.50, 3.30, 2.99 and 2.68 nmol / mL, (P <0.01). The activities of SOD and GSH were 24.16,24.67,25.56,26.02 U / mL and 0.334,0.364,0.445,0.449μmol / mg prot were higher than those in the formalin-induced injury group (P <0.01). The activity of NF-κB in the tea polyphenols groups decreased significantly compared with the formaldehyde group (P <0.01). Conclusion Tea polyphenols can increase the activity of antioxidant enzymes in endothelial cells and reduce the oxidative damage caused by formaldehyde.
