[目的]为了解释有机磷农药—甲基对硫磷对牛血清白蛋白(BSA)的作用与致毒机理。[方法]采用紫外和荧光光谱法研究甲基对硫磷与BSA的相互作用。[结论]甲基对硫磷能与BSA相互作用,其作用程度和方式与浓度、酸度和离子强度等因素有关。甲基对硫磷对BSA的荧光猝灭方式为静态猝灭,通过计算得到结合位点数(n)和结合常数(K),且结合距离(r)小于7 nm。二者的作用力为氢键或范德华力。甲基对硫磷使BSA构象发生变化,且对酪氨酸残基的影响较大。
[Objective] The aim was to explain the effect and mechanism of organophosphorus pesticide methyl parathion on bovine serum albumin (BSA). [Method] The interaction between methyl parathion and BSA was studied by ultraviolet and fluorescence spectroscopy. [Conclusion] Methyl parathion interacted with BSA, and the degree and mode of action was related to the concentration, acidity and ionic strength. The fluorescence quenching of BSA by methyl parathion is static quenching. The number of binding sites (n) and binding constant (K) were calculated and the binding distance (r) was less than 7 nm. The two forces are hydrogen bonds or van der Waals forces. Methyl parathion changes the conformation of BSA and has a greater effect on tyrosine residues.
