目的评估小剂量(低于无明显作用水平的浓度)玉米赤霉烯酮(zearalenone,ZEA)暴露后小鼠精子发生及精液质量的变化趋势,探讨小剂量ZEA暴露对雄性小鼠生殖能力的影响。方法 90只出生后21 d的雄性CD-1小鼠按随机数字表法分成3组,分别暴露于0、20、40μg/(kg·d)ZEA,处理14、28、42 d后,采用计算机辅助精液质量分析观察精子活力参数,吉姆萨染色观测精子畸形率,测量睾丸体积及质量,免疫组化染色检测生精细胞计数。结果处理42 d后,20、40μg/(kg·d)组精子活动率低于0μg/(kg·d)组[(82.80±4.34)%、(74.90±6.70)%vs(93.20±1.66)%,P=0.004、0.002];20、40μg/(kg·d)组小鼠畸形精子率高于0μg/(kg·d)组[(29.40±1.09)%、(40.80±6.17)%vs(19.40±0.45)%,P<0.001,P=0.002];20、40μg/(kg·d)组小鼠的单位面积生精细胞计数低于0μg/(kg·d)组(2 422.7±206.1、1 855.8±105.3 vs 3 189.8±306.0,P=0.013、0.001)。结论小剂量ZEA的暴露能够降低雄性小鼠的精子发生及精液质量,损害其生殖能力。
Objective To evaluate the changes of spermatogenesis and sperm quality of mice exposed to zearalenone (ZEA) at low dose (lower than the concentration without significant effect) and to explore the effect of low dose ZEA exposure on reproductive capacity of male mice . Methods Totally 90 male CD-1 mice 21 days after birth were divided into 3 groups according to the random number table, and were exposed to 0, 20, 40μg / (kg · d) ZEA, respectively. After 14, 28 and 42 days of treatment, Auxiliary semen quality analysis of sperm motility parameters, Giemsa staining sperm deformity rate, testicular volume and quality, immunohistochemical staining sperm count. Results After 42 days of treatment, the sperm motility of 20,40μg / (kg · d) group was lower than that of 0μg / (kg · d) [(82.80 ± 4.34)%, (74.90 ± 6.70)% vs (93.20 ± 1.66% , P = 0.004,0.002]. The rate of abnormal sperm in 20,40μg / (kg · d) group was higher than that in 0μg / (kg · d) group [(29.40 ± 1.09)%, (40.80 ± 6.17)% vs ± 0.45)%, P <0.001, P = 0.002]. The spermatogenic cell counts per unit area of mice in 20, 40μg / (kg · d) group were lower than those in 0μg / (kg · d) 855.8 ± 105.3 vs 3 189.8 ± 306.0, P = 0.013, 0.001). Conclusion Exposure of small doses of ZEA can reduce sperm production and sperm quality in male mice and impair their reproductive capacity.
