目的探索一种以正丁醇替代无水乙醇和二甲苯制作不同胎龄大鼠全胚切片的简便方法。方法收集胚龄14d(E14)、E16、E18、E20大鼠胚胎,固定及低浓度乙醇脱水后,部分标本按传统方法用无水乙醇脱水和二甲苯透明(无水乙醇-二甲苯法),另一部分标本用正丁醇脱水、透明(正丁醇法)。所有标本进行石蜡包埋、切片、HE染色。对两种方法从蜡块质量和切片质量等方面进行比较和评价。结果正丁醇法大大简化了操作步骤,标本处理时间容易控制,包埋块的切割性能及对肝脏组织的保存优于无水乙醇-二甲苯制片法。结论正丁醇法步骤简单且制片安全可靠,可替代无水乙醇-二甲苯法制作大鼠全胚切片。
Objective To explore a simple and convenient method for preparing whole embryos of different gestational age rats by using n-butanol instead of absolute ethanol and xylene. Methods The embryos of E14, E16, E18 and E20 embryos were collected, and after dehydration fixed and low concentration of ethanol, some samples were dehydrated by anhydrous ethanol and purified by xylene (anhydrous ethanol - xylene) according to the traditional method. The other part of the sample with n-butanol dehydration, transparent (butanol method). All specimens were paraffin-embedded, sectioned and HE stained. The two methods are compared and evaluated from the aspects of wax block quality and slice quality. Results The n-butanol method greatly simplified the operation procedure, and the sample processing time was easy to control. The cutting performance of the embedded block and the preservation of the liver tissue were better than that of the anhydrous ethanol-xylene production method. Conclusion The n-butanol method has the advantages of simple steps and safe and reliable preparation. It can replace whole-brain slices of rats by ethanol-xylene method.
