目的考察不同产地知母饮片中菝葜皂苷元及芒果苷的含量。方法采用高效液相色谱法分别测定不同产地知母饮片中菝葜皂苷元及芒果苷的含量。测定菝葜皂苷元以甲醇-水(95∶5)为流动相;流速为1.0 mL/min;柱温30℃;用蒸发光散色检测器检测,漂移管温度85℃。测定芒果苷以水-乙腈-0.85%磷酸(100∶25∶5)为流动相;流速为0.6 mL/min;柱温30℃;检测波长为258 nm。结果河北、内蒙、山西、陕西、安徽等产地菝葜皂苷元含量为1.5%~2.2%,芒果苷含量为0.3%~1.9%。结论本法测定知母饮片中菝葜皂苷元及芒果苷含量简单、快速、准确、重现性好,符合含量测定建立的要求,可以作为判断知母饮片质量的方法之一。
Objective To investigate the content of Smilagenin and mangiferin in Zhimu decoction pieces from different areas. Methods The contents of Smilagenin and mangiferin in Zhimu decoction of different areas were determined by high performance liquid chromatography. Determination of sapogenin with methanol - water (95: 5) as the mobile phase; flow rate of 1.0 mL / min; column temperature 30 ℃; with evaporative light scattering detector detection, the drift tube temperature of 85 ℃. Determination of mangiferin with water - acetonitrile-0.85% phosphoric acid (100: 25: 5) as the mobile phase; flow rate of 0.6 mL / min; column temperature 30 ℃; detection wavelength of 258 nm. Results The contents of Smilagenin in Hebei, Inner Mongolia, Shanxi, Shaanxi and Anhui originated from 1.5% to 2.2% and the content of mangiferin from 0.3% to 1.9%. Conclusion This method is simple, rapid, accurate and reproducible for the determination of Smilagenin and Mangiferin in Zhimu decoction. It can be used as a method to determine the quality of Zhimu decoction pieces.
