目的:总结近年来肿瘤生物治疗中免疫活性细胞体外增殖活化的研究进展。方法:应用PubMed及CNKI期刊全文数据库以肿瘤生物治疗、树突状细胞、细胞因子诱导的杀伤细胞、细胞培养为关键词,检索2000-01-2008-04相关树突状细胞、细胞因子诱导的杀伤细胞的文献,对资料进行初选,并查看每篇文献后的引文,以文献内容与肿瘤生物治疗、树突状细胞及细胞因子诱导的杀伤细胞培养方法相关为纳入标准。初选有关数百篇关于树突状细胞及细胞因子诱导的杀伤细胞培养方面的文章,根据纳入标准,精选50篇,最后纳入分析31篇文献。结果:树突状细胞和细胞因子诱导的杀伤细胞是进行肿瘤生物治疗的主要免疫活性细胞,它们增殖活化是成功进行生物治疗的关键。传统体外增殖活化方法发展较为成熟,新活化增殖方法是值得进一步研究的领域。结论:探索价格低廉的细胞因子替代物,对于普及生物治疗,降低成本具有十分重要的意义。
OBJECTIVE: To summarize the research progress of proliferation and activation of immunocompetent cells in tumor biotherapy in recent years. METHODS: Using the key words of tumor biotherapy, dendritic cells, cytokine-induced killer cells and cell culture, PubMed and CNKI journals were used to search the related dendritic cells from 2000-01 to 2008-04. Cytokine-induced Killer cells, primaries in data, and citations after each article are included in the literature as relevant to tumor biotherapy, dendritic cells and cytokine-induced killer cell culture. Primary selection Hundreds of articles on dendritic cells and cytokine-induced killer cell culture were selected based on inclusion criteria, 50 were selected and finally 31 were included in the analysis. RESULTS: Dendritic cells and cytokine-induced killer cells were the major immunocompetent cells for tumor biotherapy and their proliferation and activation were the key to successful biotherapy. Traditional methods of in vitro proliferation and activation of the more mature, new activation and proliferation method is a field worthy of further study. CONCLUSION: It is of great importance to explore inexpensive cytokine substitutes for universal biological treatment and cost reduction.
