目的:采用动态优化方法优选人参中人参皂苷提取工艺。方法:采用HPLC测定人参中人参皂苷Rg1,人参皂苷Re和人参皂苷Rb1的含量变化,Diamonsil RP-18e色谱柱(4.6 mm×250 mm,5μm),流动相乙腈(A)-水(B)梯度洗脱(0~7 min,16%~18%A;7~27 min,18%~30%A),流速3.0 m L·min-1,柱温35℃,DAD检测波长203 nm,进样量20μL。通过比较动态优化工艺与文献提取工艺所得人参提取物中浸膏得率、指标成分含量和指纹图谱的相似度,评价动态优化方法应用于人参中人参皂苷提取工艺优化的可行性。结果:动态优化提取工艺为8倍量70%乙醇提取3次,提取时间为第1次40 min,第2次20 min,第3次40 min,总提取物的浸膏得率为41.3 mg·g-1。动态优化提取工艺与文献提取工艺的提取物中指标成分含量及浸膏得率的RSD均<5%,指纹图谱相似度为100%,表明2种提取方法的提取物基本一致,但是动态优化提取工艺提取时间节约63%,耗电量节约60%。结论:动态优化方法可以用于人参中人参皂苷提取的工业化生产,该工艺能大幅减少提取时间,缩短生产周期,降低能耗,能够产生明显的经济价值,具有很好的应用前景。
Objective: To optimize the extraction process of ginsenosides from ginseng by dynamic optimization method. Methods: The contents of ginsenoside Rg1, ginsenoside Re and ginsenoside Rb1 in ginseng were determined by HPLC. The gradient of Diamonsil RP-18e column (4.6 mm × 250 mm, 5 μm), mobile phase acetonitrile-water (B) (0 ~ 7 min, 16% ~ 18% A; 7 ~ 27 min, 18% ~ 30% A) at a flow rate of 3.0 mL · min- Amount of 20 μL. The feasibility of using dynamic optimization method to optimize ginseng saponin extraction process was evaluated by comparing the extract yield, index components and the similarity of fingerprints of ginseng extracts obtained by dynamic optimization and literature extraction. Results: The dynamic extraction process was extracted with 8 times 70% ethanol 3 times, the extraction time was the first 40 min, the second 20 min, the third 40 min, the total extract yield was 41.3 mg · g-1. The RSDs of extract content and extract yield of active optimized extraction and literature extraction were all less than 5%, and the similarity of fingerprint was 100%, indicating that the extracts of the two extraction methods were basically the same, but the dynamic optimization extraction Process extraction time savings of 63%, power consumption savings of 60%. Conclusion: The dynamic optimization method can be used for the industrial production of ginsenosides extracted from ginseng. The process can significantly reduce the extraction time, shorten the production cycle, reduce energy consumption, and have obvious economic value, which has a good application prospect.
