目的 了解各种小儿肿瘤分子病理学特征及增殖活性, 为临床早期诊断制定合理的化疗方案及估计预后提供实验依据。方法 用流式细胞术及免疫组织化学测定100 例近5 年各类小儿肿瘤石蜡标本DNA含量及Ki67 的表达。结果 DNA含量、Ki67 表达与组织学分级呈明显的线性关系(P< 0.05) ,DNA 含量越高,Ki67 表达越高, 组织学分级越高。结论 DNA 含量测定在小儿实体恶性肿瘤的诊断中具有重要意义,DNA含量及Ki67 可独立作为小儿肿瘤病理学指标判断肿瘤恶性程度及预后, 并为化疗方案的设计提供资料。
Objective To understand the molecular pathological features and proliferative activity of various kinds of pediatric tumors and provide experimental evidence for establishing a reasonable chemotherapy regimen and estimating the prognosis in early clinical diagnosis. Methods Flow cytometry and immunohistochemistry were used to detect the DNA content and the expression of Ki67 in 100 paraffin-embedded specimens of various types of children in recent five years. Results DNA content, Ki 67 expression and histological grade showed a significant linear relationship (P <0.05), the higher the DNA content, the higher the expression of Ki 67, the higher the histological grade. Conclusion The determination of DNA content in the diagnosis of pediatric solid malignancies is of great significance, DNA content and Ki 67 can independently as a pediatric tumor pathological indicators to determine the degree of malignancy and prognosis, and to provide information for the design of chemotherapy.
