目的观察帕罗西汀对心理应激大鼠海马5-HT及c-fos表达的影响,以揭示帕罗西汀治疗心理应激引起焦虑的中枢作用机制。方法建立大鼠心理应激模型,观察各组旷场行为和海马5-HT、c-fos的表达。结果与正常对照组比较,各应激组大鼠旷场活动明显增多,海马c-fos表达显著增加,5-HT表达减少,而帕罗西汀给药能减少应激大鼠旷场活动,下调海马c-fos表达,增加5-HT表达。结论帕罗西汀可能是通过下调海马c-fos的表达,增加了海马5-HT表达,保护了海马的功能,从而发挥了中枢抗焦虑作用。
Objective To observe the effect of paroxetine on the expression of 5-HT and c-fos in hippocampus of psychological stress rats to reveal the central mechanism of paroxetine in the treatment of psychological stress-induced anxiety. Methods The rat psychological stress model was established and the behavior of open field and the expression of 5-HT and c-fos in the hippocampus were observed. Results Compared with the normal control group, the open-field activity of rats in each stress group was significantly increased, the expression of c-fos in hippocampus was significantly increased and the expression of 5-HT was decreased. Paroxetine administration reduced the open field activity in stress rats and decreased the hippocampus c-fos expression, increased 5-HT expression. Conclusion Paroxetine may play an central role in anti-anxiety by down-regulating c-fos expression in the hippocampus, increasing hippocampal 5-HT expression and protecting the function of the hippocampus.
