目的:探讨黄芪对压力损伤的兔纤维环细胞增殖及凋亡的影响。方法:藻酸盐凝胶培养兔纤维环细胞,以1Mpa持续气压静压力诱导纤维环细胞损伤,在培养基内分别加入10,50,100,500,1000mg.L-1的黄芪注射液,培养48h后CCK-8法检测细胞增殖情况,AnnexinV-PI染色检测细胞凋亡情况。结果:(1)CCK-8显示,各浓度的黄芪注射液能促进纤维环细胞的增殖,其中50mg.L-1黄芪注射液的作用最显著(与空白对照相比,P<0.01);(2)AnnexinV-PI染色显示,50mg.L-1黄芪注射液降低纤维环细胞凋亡率的作用最为显著(与空白对照相比,P<0.01),但当浓度增加至1000mg.L-1时,凋亡率虽较空白对照有所降低,但差异无显著性(P>0.05)。结论:黄芪注射液对损伤的纤维环细胞有促进增殖和抑制凋亡作用,具备用于纤维环细胞保护的潜力。
Objective: To investigate the effect of Radix Astragali on proliferation and apoptosis of rabbit fibrocystic ring cells under pressured injury. Methods: Rabbit fibrocystic ring cells were cultured with alginate gel, pressure injury of fibrocytes was induced by continuous pressure of 1 Mpa, Astragalus membranaceus injection of 10, 50, 100, 500 and 1000 mg.L-1 respectively was added into culture medium. After cultured for 48 h, CCK- 8 method to detect cell proliferation, AnnexinV-PI staining to detect apoptosis. The results showed that: (1) CCK-8 showed that Astragalus injection at various concentrations could promote the proliferation of fibrocystic ring cells, among which Astragalus injection of 50mg.L-1 had the most significant effect (P <0.01 compared with the blank control); 2) AnnexinV-PI staining showed that the effect of Astragalus injection 50mg.L-1 was most significant (P <0.01 compared with the blank control), but when the concentration was increased to 1000mg.L-1 , While the apoptosis rate was lower than the blank control, but the difference was not significant (P> 0.05). Conclusion: Astragalus injection can promote the proliferation and inhibit the apoptosis of injured fibrocystic ring cells, and has the potential of protecting the fibrocytes.
