目的研究在慢性移植肾肾病(CAN)中骨化三醇对转化生长因子-β1(TGF-β1)表达的影响及意义。方法SD大鼠到Wistar大鼠进行同种肾移植制作CAN实验大鼠模型,并随机分为2组。治疗组大鼠给予隔日腹腔注射骨化三醇5μg/kg,对照组大鼠给予等次等量的花生油。分别于术后4,8,12周分批处死动物,观察移植肾组织的病理学改变及血肌酐水平变化,用原位杂交技术检测移植肾组织中TGF-β1mRNA的变化。结果术后慢性移植肾病组均呈现CAN的病理改变,对照组的病变程度较治疗组重。移植后治疗组血清肌酐水平及TGF-β1mRNA的表达均明显低于对照组(P<0.01和0.05)。结论发生CAN时,骨化三醇会下调TGF-β1的表达,影响TGF-β1/Smads通路,减轻其病理损害。
Objective To investigate the effect of calcitriol on the expression of transforming growth factor-β1 (TGF-β1) in chronic allograft nephropathy (CAN) and its significance. Methods SD rats were Wistar rats were allotransplantation to produce CAN experimental rat model and were randomly divided into two groups. Rats in the treatment group were given intraperitoneal injection of calcitriol 5μg / kg every other day while those in the control group were given the same amount of peanut oil. The rats were sacrificed at 4, 8, and 12 weeks after operation respectively. The pathological changes and serum creatinine level were observed. The changes of TGF-β1 mRNA in renal allograft tissues were detected by in situ hybridization. Results Postoperative chronic allograft nephropathy group showed pathological changes of CAN, the control group of lesions than the treatment group. Serum creatinine and TGF-β1 mRNA expression in the post-transplantation treatment group were significantly lower than those in the control group (P <0.01 and 0.05). Conclusions In the occurrence of CAN, calcitriol down-regulates the expression of TGF-β1, and affects the TGF-β1 / Smads pathway and alleviates the pathological damage.
