目的通过运动干预高脂饮食诱导的肥胖C57/BL6小鼠,探讨运动对肥胖病理变化影响中的miRNA-33调控机制。方法 4周龄雄性C57/BL6小鼠分为3个组,对照组(6只)喂养普通饲料16周,肥胖组(8只)喂养高脂饲料16周,运动组(8只)在喂养高脂饲料8周后开始跑步8周。每周监测体重变化。三组动物处死后取血清和肝组织,用自动血生化分析仪检测血脂水平。qPCR检测肝组织miRNAs的表达情况,蛋白印记(Western-blot)检测肝组织ABCA1蛋白的表达。结果肥胖组总胆固醇为(2.26±0.28)mmol/L,高于对照组的(1.52±0.05)mmol/L和运动组的(1.87±0.21)mmol/L,差异均有统计学意义(P<0.05);肥胖组甘油三酯为(5.38±1.07)mmol/L,高于对照组的(4.15±0.64)mmol/L和运动组的(4.38±0.45)mmol/L,差异均有统计学意义(P=0.028 6、0.028 8);与对照组和运动组相比,肥胖组肝组织miRNA-33的表达降低,差异有统计学意义(P=0.009 8、0.025 9),ABCAl蛋白表达升高。结论在运动调节高脂饮食诱导的肥胖中,miRNA-33可能在其中扮演着重要角色。
OBJECTIVE: To investigate the mechanism of miRNA-33 in exercise-induced obesity pathological changes through exercise intervention in obese C57 / BL6 mice induced by high-fat diet. Methods Four-week-old male C57 / BL6 mice were divided into three groups: the control group (6 mice) fed normal diet for 16 weeks, the obese group (8 mice) fed high-fat diet for 16 weeks and the exercise group Eight weeks after the fat feed began running for eight weeks. Weekly monitoring of body weight changes. Three groups of animals sacrificed after taking serum and liver tissue, with automatic blood biochemical analyzer to detect blood lipid levels. qPCR was used to detect the expression of miRNAs in liver tissue. The expression of ABCA1 protein in liver tissue was detected by Western-blot. Results The total cholesterol in obese group was (2.26 ± 0.28) mmol / L, which was significantly higher than that in control group (1.52 ± 0.05) mmol / L and 1.87 ± 0.21 mmol / L in exercise group (P < 0.05). The level of triglyceride in obesity group was (5.38 ± 1.07) mmol / L, higher than that in control group (4.15 ± 0.64) mmol / L and 4.38 ± 0.45 mmol / L in exercise group (P = 0.028 6,0.028 8). Compared with control group and exercise group, the expression of miRNA-33 in obese group was lower than that in control group (P = 0.009 8,0.025 9) . Conclusion miRNA-33 may play an important role in the exercise-induced obesity induced by high-fat diet.
