[目的]探讨微小RNA-143(mi R-143)在结直肠癌组织中的表达及临床意义,以及上调mi R-143对于结直肠癌细胞凋亡的影响。[方法]采用实时荧光定量聚合酶链反应(real-time quantitative reverse transcription-PCR,real-time PCR)的方法在45例结直肠癌患者组织标本中检测mi R-143的表达水平,结合病理资料分析其临床意义。HCT-116细胞转染mi R-143 mimics后,用流式细胞术检测mi R-143对凋亡的影响。[结果 ]mi R-143在结肠癌组织中的相对表达量为0.48(0.24~0.91),在癌旁组织中的表达量为1.64(0.83~1.87),差异有统计学意义(P<0.001),且在黏液癌患者中下调更明显(P<0.05)。流式细胞术检测发现,与对照组的细胞凋亡率(20.00%±2.00%)相比,实验组的细胞凋亡率升高至31.83%±2.02%,差异有统计学意义(P=0.002)。[结论]mi R-143在结直肠癌组织中低表达,并且与组织学类型有关;mi R-143表达上调可促进HCT-116细胞凋亡;mi R-143具有潜在抑癌作用。
[Objective] To investigate the expression and clinical significance of micro RNA-143 (mi R-143) in colorectal cancer, and to elucidate the effect of mi R-143 on the apoptosis of colorectal cancer cells. [Methods] The expression level of mi R-143 was detected in 45 specimens of colorectal cancer patients by real-time quantitative RT-PCR (real-time PCR) combined with pathological data. Analyze its clinical significance. After miR-143 mimics were transfected by HCT-116 cells, the effect of mi R-143 on apoptosis was detected by flow cytometry. [Results] The relative expression level of mi R-143 in colon cancer tissues was 0.48 (0.24~0.91), and the expression level of mi R-143 in paracancer tissues was 1.64 (0.83~1.87). The difference was statistically significant (P<0.001). And it was significantly down-regulated in patients with mucinous carcinoma (P<0.05). Flow cytometry showed that the apoptosis rate of the experimental group was increased to 31.83%±2.02% compared with the control group (20.00%±2.00%), and the difference was statistically significant (P=0.002). ). [Conclusion] miR-143 is lowly expressed in colorectal cancer tissues and is associated with histological types. Up-regulation of miR-143 may promote apoptosis of HCT-116 cells; miR-143 has a potential anti-cancer effect.
