目的:探讨胰岛因子1(Islet1,Isl-1)在上颌牙牙髓神经生长发育的3个不同时期中投射表达的影响。方法:用Wnt1~(cre/+)雄鼠与Isl1~(f/f)雌鼠配出Wnt1~(cre/+);Isl1~(-/-)基因敲除的小鼠,做基因鉴定(PCR)。取发育不同时期第13.5d、14.5 d、15.5 d小鼠胚胎头部(实验组),同窝非基因敲除小鼠为对照,冰冻切片。用免疫荧光方法标记牙髓神经,比较第13.5 d、14.5 d、15.5 d小鼠上颌牙胚在基因敲除小鼠中与同窝对照小鼠中神经分布情况。结果:PCR结果鉴定为基因敲除小鼠以及同窝对照小鼠;分别在第13.5 d、14.5 d、15.5 d观察基因敲除小鼠与同窝对照小鼠牙髓神经,且各个时期的实验组中神经分布明显减少,对照组中神经分布较多。结论:在牙髓神经的生长发育过程中,Isl-1这一转录因子对牙髓神经的投射表达产生影响,可能是Isl-1调控牙胚周围的转录因子,并影响牙髓神经的投射。
Objective: To investigate the effect of Islet1 (Isl-1) on the projection of maxillary teeth in three different periods of development of dental pulp nerve. Methods: The Wnt1 ~ (cre / +); Isl1 ~ (- / -) knockout mice were assigned to Wnt1 ~ (cre / +) male mice and Isl1 ~ (f / f) PCR). The embryonic heads (experimental group), littermate non-gene knockout mice on the 13.5d, 14.5d and 15.5d developmental stages were used as control and frozen sections. The dental pulp nerve was labeled by immunofluorescence and the distribution of nerve in the knockout mice and littermate control mice on the day 13.5, 14.5 and 15.5 days were compared. Results: The results of PCR were identified as knockout mice and littermate control mice. The dental pulp nerve of gene knockout mice and littermate control mice were observed on day 13.5, 14.5 and 15.5, and the experiment of each period Nerve distribution in the group was significantly reduced in the control group more nerve distribution. CONCLUSIONS: Isl-1 transcription factor affects the projection of dental pulp during the growth and development of dental pulp nerve. It may be that Isl-1 regulates the transcription factors around tooth germ and affects the projection of dental pulp nerve.
