目的:优选微波提取板蓝根中木脂素部位的工艺。方法:以总木脂素含量及clemastanin B(CB)单体含量作为筛选指标,分别应用紫外分光光度法、HPLC对两者进行含量测定,采用正交实验法对提取过程中的微波辐照功率、提取时间、料液比、浸泡时间4个因素进行优选。结果:最佳提取条件为辐照功率700 W,加15倍量水提取2次,每次8 min,提取前浸泡15min,在此条件下所得总木脂素和CB的提取率分别达到3.39%,0.019 3%,与回流提取3.5 h的提取率无显著差异。结论:本方法具有快速、高效、方便、节能等优点,值得推广应用。
Objective: To optimize microwave extraction technology of lignans in Radix isatidis. Methods: Total lignin content and monomer content of clemastanin B (CB) were used as screening indexes. The content of clemastanin B (CB) was determined by UV spectrophotometry and HPLC respectively. The orthogonal experiment was used to determine the microwave irradiation power , Extraction time, solid-liquid ratio, soaking time 4 factors are preferred. Results: The optimal extraction conditions were as follows: the irradiation power 700 W, 15 times the amount of water extraction 2 times, each time 8 min, 15 min before extraction, the extraction rate of total lignans and CB under this condition reached 3.39% , 0.019 3%, respectively. There was no significant difference between the extraction rate and the reflux extraction time of 3.5 h. Conclusion: The method is fast, efficient, convenient, energy saving, etc., it is worth promoting.
