目的探讨丹参多酚酸盐对电离辐射导致的人脐静脉内皮细胞(HUVEC)系ECV-304损伤的影响。方法用不同浓度丹参多酚酸盐0、0.5、1.0mg/L处理ECV-304细胞后,分别给予8、16Gy 6MV-X射线照射24h。采用黄嘌呤氧化酶法和硫代巴比妥酸法分别检测SOD活性和MDA含量,流式细胞术检测细胞凋亡率。结果丹参多酚酸盐呈浓度依赖性地增加受辐射损伤ECV-304细胞中SOD活性,降低MDA含量,减少细胞凋亡率。结论丹参多酚酸盐对辐射造成的血管内皮细胞损伤具有明显的保护作用。
Objective To investigate the effects of Salvianolic acid polyphenols on the injury of human umbilical vein endothelial cells (HUVEC) line ECV-304 induced by ionizing radiation. Methods ECV-304 cells were treated with Salviae miltiorrhizae polyphenols at different concentrations (0, 0.5 and 1.0 mg / L) for 8 h and 6 Gy 6MV-X irradiation for 24 h. The activity of SOD and MDA content were detected by xanthine oxidase method and thiobarbituric acid method respectively. The apoptosis rate was detected by flow cytometry. Results Salviae miltiorrhiza polyphenols could increase the SOD activity in ECV-304 cells in a dose-dependent manner, decrease the MDA content and decrease the apoptosis rate. Conclusion Radix salviae miltiorrhizae polyphenols have obvious protective effect on the injury of vascular endothelial cell induced by radiation.
