支原体基因组全DNA分子量较小,本研究直接制备肺炎支原体(MF),人型支原体(MH)和解脲脲原体(UU)中3个型SU1,SU4,SU8全DNA光敏生物素探针,用斑点杂交鉴定表明探针可用于检测同种支原体,但3个型UU探针不能用于型别鉴定。探针检测灵敏度较一般片段DNA探针灵敏度低,可能是由于前者分子量放大,降低了杂交速率和效率。直接提取全DNA作光敏生物素标记,简化了探针制备过程,且无放射性损害及衰减,探针易于保存,可供一般实验室及化验室使用
Mycoplasma genome DNA molecular weight is small, the study directly prepared three Mycoplasma pneumoniae (MF), Mycoplasma hominis (MH) and Uu UU three SU1, SU4, SU8 all-optical biotin probe with Dot blot hybridization assays indicate that the probe can be used to detect allogeneic mycoplasma, but 3-type UU probes can not be used for type identification. The detection sensitivity of the probe is lower than the sensitivity of the general fragment DNA probe, probably due to the amplification of the former molecular weight and the decrease of the hybridization rate and the efficiency. Direct extraction of whole DNA for photobiotin labeling simplifies probe preparation, and no radioactive damage and attenuation, the probe is easy to save for general laboratory and laboratory use
