目的比较实时荧光PCR检测(FQ-PCR)与RT-PCR在鉴定甲型流感病毒中的优劣,建立能快速简便、特异地从临床标本中检测并鉴定流感病毒的新方法。方法对200份临床采集的咽拭子样本进行FQ-PCR与RT-PCR检测,综合比较2种监测方法之间的差异。结果 FQ-PCR检测200份临床咽拭标本,96份甲型阳性,阳性率为48.0%;从RNA提取到检测结果仅需3~3.5 h。RT-PCR检测200份临床咽拭标本,69份阳性,阳性率为34.5%;从RNA提取到检测结果约需8 h。结论 FQ-PCR方法快速、特异、灵敏,在甲型流感病毒检测中具有较大的应用价值。
Objective To compare the advantages and disadvantages of real-time fluorescent PCR (FQ-PCR) and RT-PCR in the identification of influenza A virus and to establish a new method that can detect and identify influenza virus from clinical samples quickly and easily. Methods FQ-PCR and RT-PCR were performed on 200 clinically collected throat swab samples, and the differences between the two monitoring methods were compared. Results FQ-PCR detection of 200 clinical pharyngeal swab specimens, 96 were positive for type A, the positive rate was 48.0%; extracted from the RNA to the test results only 3 ~ 3.5 h. Ninety-six positive swab samples were detected by RT-PCR, with a positive rate of 34.5%. The RNA extraction results were about 8 hours. Conclusion FQ-PCR method is rapid, specific and sensitive and has great value in the detection of influenza A virus.
