目的 探讨人体恶性肿瘤裸鼠移植瘤药敏试验与PCNA表达的内在联系。方法 新鲜实体肿瘤裸鼠肾包膜下移植 3 0例 ,每例 2 4只BALB c裸鼠 ,随机分为 8组 (有 10例只分为 5或 6组 ) ,每组 2只腹腔注射 1种抗癌药 ,1只注射生理盐水对照 ;测定药敏结果后移植瘤行组织学和PCNA检测。结果 移植瘤 12 2 8个 ,其中对照组 40 0个 ,呈正生长60 3 % ,无生长 2 0 8% ,负生长 18 9% ;检测抗癌药 13种 ,敏感 5 1次 ,较敏感 5 2次 ,不敏感 90次 ;送检实验后移植瘤 2 88只 ,光镜下瘤细胞可见率 75 3 % ;PCNA表达阳性率在原始肿瘤标本与对照组中为 69 2 %与 70 8% (P >0 0 5 ) ,在原始肿瘤标本与药敏组中为 69 2 %与 3 9 8% (P <0 0 5 )。结论 人体恶性肿瘤裸鼠移植瘤能保持原肿瘤的生物特性 ;移植瘤药敏组间以及与对照组比较 ,PCNA的免疫标记差异有显著性。提示瘤体对药物敏感度与PCNA呈负相关 ;PCNA指数也与预后有关。
Objective To investigate the relationship between susceptibility test and PCNA expression in human malignant tumor xenografts in nude mice. Methods Thirty (40) BALB / c nude mice were randomly divided into 8 groups (10 cases were divided into 5 or 6 groups), 2 mice in each group were injected intraperitoneally with 1 Anti-cancer drugs, a saline injection control; determination of drug sensitivity after transplantation tumor histological and PCNA detection. RESULTS: A total of 1228 xenografts were found in the control group, of which 40 0 were positive, 60 3% positive, 28.0% non-growth, and 18.9% negative. 13 kinds of anticancer drugs were detected, 5 sensitive and 5 5 sensitive Times, insensitive to 90 times. After the test, 2 88 transplanted tumors were observed, and the tumor cell visible rate was 75 3% under light microscopy. The positive rate of PCNA expression was 69 2% and 70 8% in the original tumor specimen and the control group > 0 0 5), 69 2% and 39 8% (P 0 05) in the original tumor specimens and susceptibility groups. Conclusion The transplanted human malignant tumor in nude mice can maintain the biological characteristics of the original tumor. Compared with the sensitive group and the control group, the immuno-labeling of PCNA has significant difference. Tip tumor sensitivity to drugs and PCNA was negatively correlated; PCNA index and prognosis.
