目的观察通心络胶囊对糖尿病周围神经病变小鼠的作用。方法 KK/Upj-Ay小鼠随机分为4组,模型组与高、中、低3个剂量通心络组(0.8,0.4,0.2 g·kg-1),另设C57BL/6小鼠为对照组。灌胃给药12周,测定运动神经传导速度(MNCV)和感觉神经传导速度(SNCV);实时定量荧光聚合酶链反应和蛋白质印迹法测定坐骨神经肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)、白介素-1β(IL-1β)和核因子-κB(NF-κB)表达。结果与模型组比较,通心络组的小鼠MNCV、SNCV明显增快(P<0.05,P<0.01),TNF-α、IL-6、IL-1β和NF-κB的表达显著下降(P<0.05,P<0.01)。结论通心络胶囊可抑制NF-κB通路,抑制糖尿病周围神经病变小鼠炎症因子表达,从而改善糖尿病周围神经病变。
Objective To observe the effect of Tongxinluo Capsule on diabetic peripheral neuropathy in mice. Methods KK / Upj-Ay mice were randomly divided into 4 groups: model group and high, medium and low dose Tongxinluo group (0.8,0.4,0.2 g · kg-1), another C57BL / 6 mice were Control group. MNCV and SNCV were measured by intragastric administration for 12 weeks. The expressions of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-10) in sciatic nerve were detected by real- time quantitative polymerase chain reaction and Western blotting. 6 (IL-6), interleukin-1β (IL-1β) and nuclear factor-κB (NF-κB). Results Compared with the model group, MNCV and SNCV in Tongxinluo group were significantly increased (P <0.05, P <0.01), and the expression of TNF-α, IL-6, IL- <0.05, P <0.01). Conclusion Tongxinluo Capsule can inhibit the NF-κB pathway and inhibit the expression of inflammatory cytokines in diabetic peripheral neuropathy mice, so as to improve diabetic peripheral neuropathy.
