目的:探讨高糖状态对体外培养血管内皮细胞迁移、增殖能力的影响,以及其潜在可能的分子机理。方法:通过不同质量分数的葡萄糖干预血管内皮细胞后,用CCK-8评估内皮细胞增殖情况;用transwell实验检测内皮细胞的迁移能力;用Real-time PCR检测腺苷酸环化酶(adenylate cyclase,AC)mRNA的表达;用Western blotting检测PKA、pPKA蛋白的表达情况。结果:高质量分数葡萄糖干预血管内皮细胞后,血管内皮细胞迁移、增殖能力明显下降,同时AC mRNA表达明显下降,PKA蛋白表达无明显变化,而p-PKA蛋白表达明显下降。结论:高质量分数葡萄糖使血管内皮细胞迁移、增殖能力下降,c AMP-PKA信号通路的活化可能参与了血管内皮细胞增殖、迁移及修复功能。
Objective: To investigate the effect of high glucose on migration and proliferation of vascular endothelial cells cultured in vitro and its potential molecular mechanism. Methods: The proliferation of endothelial cells was evaluated by CCK-8 after different concentrations of glucose intervention. The transwell assay was used to detect the migration of endothelial cells. The expression of adenylate cyclase AC) mRNA expression was detected by Western blotting PKA, pPKA protein expression. Results: The migration and proliferation of vascular endothelial cells were significantly decreased after high-quality fraction glucose intervention on vascular endothelial cells. Meanwhile, the expression of AC mRNA was significantly decreased while the expression of p-PKA protein was not significantly changed. CONCLUSION: High-quality fraction glucose can induce the migration of vascular endothelial cells and decrease the proliferative capacity. Activation of c AMP-PKA signaling pathway may be involved in the proliferation, migration and repair of vascular endothelial cells.
