目的 研究蝌蚪提取液 (T871 3)对白血病细胞的作用机制。 方法 利用细胞形态学观察、细胞化学、TUNEL法等技术 ,并采用原位mRNA杂交和完整细胞原位斑点印迹技术 ,观察T871 3诱导HL 6 0细胞分化和凋亡过程中c myc、c myb、bcl 2癌基因表达的变化。 结果 1 T871 3能抑制HL 6 0细胞分裂增殖。 2 T871 3在低浓度时诱导HL 6 0细胞向单核 /巨噬样细胞方向分化 ,高浓度时诱导细胞凋亡。 3 T871 3诱导HL 6 0细胞分化的过程伴随着c myc、c myb癌基因表达的下调 ,诱导HL 6 0细胞凋亡的过程伴随着c myc、bcl 2癌基因表达的下调。 结论 T871 3可能通过调节癌基因的表达发挥其诱导HL 6 0细胞分化和凋亡的作用。
Objective To study the mechanism of action of wolfberry extract (T871 3) on leukemia cells. Methods Cellular morphology, cytochemistry, TUNEL assay and other techniques were used. In situ mRNA hybridization and in situ dot blotting of intact cells were performed to observe the differentiation and apoptosis of HL 60 cells induced by T871 3 during the process of c myc and c myb. Bcl 2 Oncogene expression changes. Results 1 T871 3 inhibited the proliferation of HL 60 cells. 2 T871 3 induces differentiation of HL 60 cells to mononuclear/macrophage-like cells at low concentrations, and induces apoptosis at high concentrations. 3 The process of differentiation induced by T871 3 in HL 60 cells was accompanied by the down-regulation of c myc and c myb oncogene expression. The induction of apoptosis in HL 60 cells was accompanied by the down-regulation of c myc and bcl 2 oncogene expression. Conclusion T871 3 may play a role in inducing differentiation and apoptosis of HL 60 cells by regulating oncogene expression.
