目的探讨同型半胱氨酸(Hcy)及其代谢关键酶蛋氨酸合成酶(methionine synthase,MS)基因多态性与颅内动脉瘤的关系。方法采用化学发光技术检测76例颅内动脉瘤患者和77例健康成人的血浆Hcy水平,并利用聚合酶链式反应技术检测其MS基因多态性。结果颅内动脉瘤组血浆Hcy水平为(16.14±6.72)μmol/L高于对照组的(11.20±3.20)μmol/L,两组间差异有统计学意义(P<0.05)。颅内动脉瘤组的MS D919G DG基因型频率较正常对照高(P<0.01),患颅内动脉瘤的风险是对照组的6.35倍。两组等位基因频率差异无统计学意义(P>0.05)。MS D919G突变基因型TC、TG、Hcy浓度分组比较结果显示,颅内动脉瘤组不同基因型TG、TC浓度差异无统计学意义(P>0.05),DG基因型Hcy浓度较其他基因型高,差异有统计学意义(P<0.05);颅内动脉瘤组Hcy浓度较对照组高,差异有统计学意义(P<0.05),但两组TG和TC水平差异无统计学意义(P>0.05);对照组不同基因型间TG、TC、Hcy水平差异无统计学意义(P>0.05)。结论高Hcy血症与颅内动脉瘤病发生有一定关系,MS D919G DG基因型与颅内动脉瘤可能有相关。
Objective To investigate the relationship between homocysteine (Hcy) and methionine synthase (MS) gene polymorphisms and intracranial aneurysms. Methods The plasma Hcy levels in 76 patients with intracranial aneurysms and 77 healthy adults were detected by chemiluminescence technique. The polymorphism of MS gene was detected by polymerase chain reaction (PCR). Results The level of Hcy in intracranial aneurysm group was (16.14 ± 6.72) μmol / L higher than that in control group (11.20 ± 3.20) μmol / L, the difference was statistically significant (P <0.05). The frequency of MS D919G DG genotype in intracranial aneurysm group was higher than that in control group (P <0.01), and the risk of intracranial aneurysm was 6.35 times of that in control group. There was no significant difference in allele frequencies between the two groups (P> 0.05). MS, D919G mutation genotype TC, TG, Hcy concentration group comparison showed that intracranial aneurysm group of different genotype TG, TC concentration was no significant difference (P> 0.05), DG genotype Hcy concentration higher than other genotypes, (P <0.05). The concentration of Hcy in intracranial aneurysm group was higher than that in control group (P <0.05), but there was no significant difference in TG and TC between the two groups (P> 0.05) ). There was no significant difference in the levels of TG, TC and Hcy among different genotypes in the control group (P> 0.05). Conclusions Hcy hyperlipidemia is related to the occurrence of intracranial aneurysm. The genotype of DG in MS D919G may be correlated with intracranial aneurysm.
