目的比较48批次同一来源不同采收期橘皮中多糖类成分随采收时间的动态积累规律,研究橘皮药材多糖类成分的积累与采收时间之间的相关性。方法采用定点、定植株、定品种的方法收集不同采收时间的橘皮,通过Sevage试剂反复除蛋白制得精制陈皮多糖,计算陈皮多糖对葡萄糖的换算因子f;运用正交试验法优选陈皮多糖最佳显色条件,采用紫外分光光度法测定不同采收时间橘皮药材中多糖类成分的含量。结果四川、重庆产的红橘皮中多糖含量的整体变化趋势随采收时间的增加而不断增加,最后呈稳定趋势,即随着橘皮药材的成熟度的增加多糖含量亦随之增加。结论该研究为橘皮药材多糖类成分的深入研究和相应采收期的制定以及橘皮药材质量标准的提升提供了一定的参考,亦为阐释青皮、陈皮“一体二用”和同源而不同效的物质基础差异提供依据。
OBJECTIVE To compare the dynamic accumulation of polysaccharides in orange peel from different sources in 48 batches of different harvest time with the harvesting time and to study the correlation between the accumulation of polysaccharides and the harvesting time in orange peel. Methods The orange peel of different harvesting time was collected by fixed point, colonization and fixed variety method. The refined peel polysaccharide was obtained by repeated protein removal by Sevage reagent. The conversion factor f of dried peel polysaccharides was calculated. The orthogonal test was used to optimize the quality of dried peel polysaccharides The optimum conditions for the color development were determined by ultraviolet spectrophotometry of different harvest time orange peel polysaccharides ingredients. Results The overall trend of polysaccharide content in the red peel of Sichuan and Chongqing increased with the increase of the harvesting time and finally showed a steady trend. That is to say, with the maturity of the orange peel, the polysaccharide content also increased. Conclusion This study provides some references for the further research on the polysaccharide composition of orange peel medicinal materials and the establishment of the corresponding harvesting period as well as the improvement of the quality standard of the orange peel medicinal materials. It also provides a reference for the improvement of the quality standards of peel orange peel, Source and not the basis of different material basis to provide the basis.
