目的 观察肝郁脾虚因素对S180荷瘤小鼠肿瘤生长和T细胞免疫功能的影响。方法 取腹腔传代第 7天的S180肿瘤细胞 ,调细胞浓度至 1× 10 10 L-1,接种于昆明小鼠右腋皮下 ,0 2mL 只。接种后当天用束缚加饮食失节的方法造成肝郁脾虚证 ,并设单纯肝郁脾虚证、单纯肿瘤和正常对照组。 10d后处死小鼠 ,剥取肿瘤称瘤重 ,取脾脏检测T淋巴细胞增殖及产生IL 2、IFN γ的能力 ,并摘取胸腺称重计算胸腺指数。结果 肝郁脾虚因素可明显增加S180荷瘤小鼠肿瘤重量 (P <0 0 1) ,降低荷瘤小鼠脾T淋巴细胞增殖和产生IL 2和IFN γ的能力 (P <0 0 1) ,并可使小鼠胸腺指数明显降低 (P <0 0 1)。结论 肝郁脾虚因素可加重抑制荷瘤小鼠的细胞免疫功能 ,对肿瘤生长有促进作用。
Objective To observe the effects of the factors of liver stagnation and spleen deficiency on tumor growth and T cell immune function in S180 tumor-bearing mice. Methods S180 tumor cells from the 7th day after abdominal passage were transferred to a concentration of 1×10 10 L −1 and inoculated into the right iliac crest of Kunming mice with 0 mL. On the day after inoculation, the symptoms of stagnation and spleen deficiency were caused by the combination of restraint and dietary disorders, and simple liver depression and spleen deficiency syndrome, simple tumors and normal control groups were set up. After 10 days, the mice were sacrificed and the weight of the tumors was removed. The spleen was used to detect the proliferation of T lymphocytes and the ability to produce IL-2 and IFN-γ. The thymus was weighed to calculate the thymus index. Results The factors of liver stagnation and spleen deficiency significantly increased the tumor weight of S180 tumor-bearing mice (P < 0.01), decreased the proliferation of splenic T lymphocytes and the production of IL 2 and IFN γ in tumor-bearing mice (P <0 01). And can significantly reduce the mouse thymus index (P <0 01). Conclusion The factors of liver depression and spleen deficiency can aggravate the cellular immune function of tumor-bearing mice and promote tumor growth.
