[目的]建立反相高效液相色谱法测定人血浆中对氯苯氧乙酸的含量。[方法]以卡马西平为内标,采用YMC-C18柱,以0.005mol.L-1醋酸铵缓冲液-乙腈溶液(pH调3.69)(67:33)为流动相,流速1.0ml.min-1,紫外检测波长为225nm。[结果]标准曲线线性范围为:50~0.3125(μg.ml-1);对氯苯氧乙酸的检测限为0.3125μg.ml-1;提取回收率为:86.78%~98.48%;方法回收率为97.87%~103.2%;日内RSD在1.77%~3.75%之间,日间RSD在1.41%~7.21%之间。[结论]该方法操作简单,结果准确,重现性好,可用于人血浆中的对氯苯氧乙酸的含量测定和药动学研究。
[Objective] To establish an RP-HPLC method for the determination of p-chlorophenoxyacetic acid in human plasma. [Method] With carbamazepine as internal standard, YMC-C18 column was used with mobile phase of 0.005mol.L-1 ammonium acetate buffer-acetonitrile (pH = 3.69) (67:33) -1, UV detection wavelength of 225nm. [Result] The linear range of standard curve was 50 ~ 0.3125 (μg.ml-1), the detection limit of p-chlorophenoxyacetic acid was 0.3125μg.ml-1, the recoveries were 86.78% -98.48% Was 97.87% ~ 103.2%. The intraday RSD was between 1.77% and 3.75%, and the intraday RSD was between 1.41% and 7.21%. [Conclusion] The method is simple, accurate and reproducible. It can be used in the determination of p-chlorophenoxyacetic acid and pharmacokinetics in human plasma.
