目的:研究土荆皮酸(PAB)对体外培养的SKOV3细胞的生物学效应和作用机制。方法:用MTT法、流式细胞术、电镜检测体外培养的SKOV3细胞在PAB作用下的增殖抑制及凋亡程度,用RT-PCR法检测P53/Bcl-2/Bax的表达水平。结果:①PAB对SKOV3细胞的增殖抑制作用随浓度升高而明显增强,其IC50约10μmol/L。②流式细胞术证实PAB呈浓度依赖性改变细胞周期分布,一方面降低G0/G1期的细胞比例,另一方面增高G2/M期细胞的比例。③RT-PCR法显示SKOV3细胞在10μmol/LPAB作用12、24、48h均显示Bax上调和Bcl-2的下调,P53则未检测到表达。结论:在体外PAB能抑制SKOV3细胞增殖,诱导其凋亡,其诱导凋亡机制可能与Bax的上调和Bc1-2的下调相关。
Objective: To study the biological effect and mechanism of soil pseudohogamic acid (PAB) on cultured SKOV3 cells. Methods: MTT assay, flow cytometry and electron microscopy were used to detect the proliferation inhibition and apoptosis of SKOV3 cells cultured in vitro under the action of PAB. The expression of P53 / Bcl-2 / Bax was detected by RT-PCR. Results: ① The inhibitory effect of PAB on proliferation of SKOV3 cells was significantly enhanced with the increase of its concentration, and its IC50 was about 10μmol / L. ② Flow cytometry confirmed that PAB changed the cell cycle distribution in a concentration-dependent manner. On the one hand, it decreased the proportion of cells in G0 / G1 phase and increased the proportion of G2 / M phase cells in the other. ③ RT-PCR showed that SKOV3 cells showed up-regulation of Bax and down-regulation of Bcl-2 at 12, 24 and 48 hours after treated with 10μmol / LPAB, but no expression of P53. CONCLUSION: PAB can inhibit proliferation and induce apoptosis of SKOV3 cells in vitro. The mechanism of apoptosis induced by PAB may be related to the up-regulation of Bax and down-regulation of Bcl-2.
