本研究用12对引物组合对杂交水稻生产中广泛应用的优良杂交组合及其亲本(共15个材料)进行了AFLP分析。通过优选,筛选出 3对引物,即 E-ACT/M·CAC, E-AAC/M-CAA及 E-AAG/M-CAA,它们是本研究中用来区分所研究的 15个材料所需的最少引物组合。然后根据所选用的16个多态性DNA条带的有无,把每个材料的DNA指纹转换成由1和0组成的指纹数据。在此基础上,开发了一套应用软件。在实际应用中,将样品的扩增结果转换成指纹数据,由计算机辅助进行种子鉴定。
In this study, 12 pairs of primer combinations were used to analyze AFLP for the excellent hybrid combinations widely used in hybrid rice production and their parents (15 materials in total). Three pairs of primers, E-ACT / M-CAC, E-AAC / M-CAA and E-AAG / M-CAA, were selected by preference and were used in this study to distinguish the 15 materials studied Minimum primer combination.
