目的:研究18β-甘草次酸长期灌胃对正常大鼠血细胞的影响。方法 :将96只Wistar大鼠随机分成正常对照组、低剂量组(25μg/g)、中剂量组(50μg/g)和高剂量组(100μg/g)4组。药物组分别给予对应剂量的甘草次酸灌胃,正常对照组给予等体积生理盐水,1 d 1次。于给药14周、22周、30周及停药4周时,检测大鼠红细胞计数(RBC)、血红蛋白含量(HGB)、红细胞压积(HCT)、平均红细胞体积(MCV)、平均红细胞血红蛋白浓度(MCHC)、平均红细胞血红蛋白含量(MCH)、白细胞数计数(WBC)、中性粒细胞比例(N%)、淋巴细胞比值(L%)、血小板计数(PLT)。结果:14周、22周及30周时甘草次酸组(低、中、高剂量)RBC、HGB、MCHC、N%、PLT较正常对照组升高,MCV、L%较正常对照组降低,但3个剂量组之间差别无统计学意义,且上述指标于停药4周时接近正常对照组(差别无统计学意义)。同一指标于14周、22周及30周各时间点之间对比,差别无统计学意义,但与停药4周时相比差别有统计学意义。14周、22周、30周及停药4周时HCT、MCH、WBC与正常对照组对比,差别无统计学意义;同一时间点各组之间对比、同一组不同时间点之间对比,差别均无统计学意义。结论:18β-甘草次酸长期灌胃可引起一过性的RBC、HGB、MCHC、N%、PLT的升高和MCV、L%的降低,这种影响在停药一定时间后消除;对HCT、MCH、WBC无明显影响。
Objective: To study the effects of long-term administration of 18β-glycyrrhetinic acid on blood cells in normal rats. Methods: Ninety - six Wistar rats were randomly divided into four groups: normal control group, low dose group (25μg / g), middle dose group (50μg / g) and high dose group (100μg / g). Drug group were given the corresponding dose of glycyrrhetinic acid gavage, normal control group given an equal volume of saline, 1 d 1 times. The RBC, HGB, HCT, MCV, RBC and RBC were measured at 14, 22, 30 and 4 weeks after drug administration. MCHC, MCH, WBC, neutrophil ratio, lymphocyte ratio, and platelet count (PLT). Results: The levels of RBC, HGB, MCHC, N% and PLT in glycyrrhetinic acid group (low, middle and high dose) were significantly higher than those in normal control group at 14, 22 and 30 weeks However, the difference between the three dose groups was not statistically significant, and the above indicators in the withdrawal of 4 weeks close to the normal control group (no statistically significant difference). The same index at 14 weeks, 22 weeks and 30 weeks between the time points, the difference was not statistically significant, but compared with 4 weeks after stopping the difference was statistically significant. HCT, MCH and WBC in 14 weeks, 22 weeks, 30 weeks and 4 weeks after drug withdrawal were not significantly different from those in control group. There was no significant difference in HCT, MCH and WBC between the two groups at the same time point No statistical significance. CONCLUSION: Long-term gavage administration of 18β-glycyrrhetinic acid can cause transient increase of RBC, HGB, MCHC, N%, PLT and decrease of MCV and L% after a period of withdrawal, , MCH, WBC no significant effect.
