目的建立一种从烟叶中提取分离茄尼醇的方法,以期为高纯度茄尼醇的生产提供可靠的技术指标。方法采用超临界CO2萃取法从烟叶中萃取得到茄尼醇粗品,通过硅胶柱色谱及结晶进行分离纯化。结果超临界CO2萃取茄尼醇的最佳实验条件为:萃取压力20MPa、萃取温度45℃、萃取时间3h。在此最佳实验条件下,茄尼醇的提取率为94.5%,纯度约为33.8%,并将此粗品经过硅胶柱色谱及后续结晶,得到纯度为93.4%的茄尼醇样品,折纯一次收率为70%。结论本方法简便可行,获得的茄尼醇纯度高、收率高。
Objective To establish a method for extracting solanesol from tobacco leaves in order to provide reliable technical indicators for the production of high-purity solanesol. Methods The crude solanesol was extracted from tobacco leaves by supercritical CO2 extraction and separated by silica gel column chromatography and crystallization. Results The optimum experimental conditions for extraction of solanesol by supercritical CO2 were: extraction pressure 20MPa, extraction temperature 45°C, and extraction time 3h. Under this optimal experimental condition, the extraction rate of solanesol was 94.5% and the purity was about 33.8%. The crude product was subjected to silica gel column chromatography and subsequent crystallization to obtain a solanesol sample with a purity of 93.4%. The yield is 70%. Conclusion The method is simple and feasible, and the obtained solanesol has high purity and high yield.
