目的:观察脾虚模型大鼠肠黏膜味蛋白(Gαgust)表达情况以及健脾方药的干预作用,探讨肠道味觉关键信号分子Gαgust异常表达与脾虚证的发病相关性。方法:采用大黄脾虚模型,并设肾虚模型对照,通过QPCR法、Western blot法、免疫组化法对脾虚治疗组、脾虚自然恢复组、脾虚模型组、肾虚对照组及正常对照组Gαgust进行检测。结果:脾虚模型组Gαgust表达与正常对照组、脾虚自然恢复组相比下降(P<0.05),尤其与肾虚组比较也有显著性差异(P<0.05),经健脾治疗后脾虚治疗组Gαgust表达比脾虚自然恢复组、脾虚模型组、肾虚对照组表达上升。结论:肠道味觉关键信号分子Gαgust异常表达与脾虚证的发病密切相关,而健脾方药对其有一定的调节作用。
Objective: To observe the expression of Gαgust in spleen deficiency model rats and the intervention of invigorating the spleen recipe, and to explore the correlation between Gαgust abnormal expression of gut taste and pathogenesis of spleen deficiency syndrome. Methods: The model of rhubarb and spleen deficiency was established, and the model of kidney deficiency was established. Gαgust was detected by QPCR, Western blot and immunohistochemistry in spleen deficiency group, spontaneous spleen deficiency group, spleen deficiency model group, kidney deficiency control group and normal control group. Results: The expression of Gαgust in spleen deficiency model group was significantly lower than that in normal control group and spleen deficiency group (P <0.05), especially in kidney deficiency group (P <0.05). After spleen deficiency treatment, Gαgust expression Compared with the spontaneous spleen recovery group, the spleen deficiency model group and the kidney deficiency control group increased. Conclusion: The abnormal expression of Gαgust, a key signaling molecule of gut taste, is closely related to the pathogenesis of spleen deficiency syndrome. However, Jianpi Recipe has some regulatory effects.
