酶分析法有较强的特异性和一定的灵敏度,已广泛用于生化物质的分析。但生物化学家对方法灵敏度的要求是永无知足的。许多重要生化物质在组织内的浓度很低。当试样量极微时,一般酶分析法的应用受到了限制。酶循环(enzymatic cycling)具有化学性放大作用,理论上可无限放大一般酶分析法的灵敏度。目前酶循环放大分析法已可准确定量10~(-15)—10~(-18)摩尔的生化物质。由于商品酶试剂的增加,酶循环放大分析法正在迅速发展。
Enzymatic analysis has strong specificity and a certain degree of sensitivity, has been widely used in biochemical analysis. However, the biochemist’s sensitivity to the method is never satisfied. Many important biochemical substances in the tissue concentrations are low. When the sample size is very small, the general application of enzyme analysis has been limited. Enzymatic cycling has the effect of chemical amplification, which in theory can amplify the sensitivity of typical enzyme assays. At present, Enzyme Cycle Amplification Analysis has been able to accurately quantify 10 ~ (-15) -10 ~ (-18) mol of biochemical substances. Due to the increase of commercial enzyme reagents, Enzyme Loop Amplification Analysis is rapidly developing.
