在我国长江流域冬油菜区,油菜小孢子培养获得再生苗后需要经历漫长的越夏过程,目前的普遍做法是多次继代培养,该方法操作繁琐、技术要求高且易污染。为解决复杂低效的越夏问题,本研究建立了水培技术体系实现油菜小孢子苗越夏过程。试验结果表明,利用水培技术油菜小孢子幼苗存活率超过90%,小孢子苗直接移栽至大田存活率超过93%。此外,该技术节约90%以上的试剂费用和劳务费用,极大地简化了小孢子苗越夏操作过程。
In China’s winter rapeseed area of the Yangtze River Basin, microspore culture of rapeseeds need to go through a long summer process after regenerating seedlings. At present, the common practice is to subculture many times. The method is cumbersome to operate, has high technical requirements and is easy to be polluted. In order to solve the complex and inefficient summer problem, this study established a hydroponic technology system to achieve the process of microspore sprouting in summer. The results showed that the survival rate of microspore seedling of rape using hydroponics was over 90%, and the survival rate of microspore seedlings transplanted to field was more than 93%. In addition, the technology saves more than 90% reagent costs and labor costs, which greatly simplifies the microspore sprout summer operation process.
