目的:筛选与优化银翘散提取工艺,为银翘散复方制剂谱效关系研究奠定基础。方法:采用正交实验筛选银翘散最佳提取条件,分别以水、30%、50%、70%及95%乙醇回流提取银翘散,以高效液相色谱法测定银翘散指纹图谱,以连翘苷、绿原酸、牛蒡苷、橙皮苷等物质的含量作为工艺评价指标,筛选与优化银翘散提取工艺。结果:提取溶剂量及提取次数是影响出银翘散膏量重要因素,其次为提取时间;高效液相色谱法测定不同银翘散提取物均含有27个峰所属化合物。结论:方法简便,重现性好,用于银翘散提取物的鉴别,具有良好的专属性;银翘散各成分之间色谱峰形良好,基本达基线分离,各提取方法的银翘散提取物的某些化学成分及含量有差异,可作为银翘散抗病毒谱效关系研究基础。
OBJECTIVE: To screen and optimize the extraction process of Yinqiao Powder and lay a foundation for the study on the relationship between the spectral properties of Yinqiaoqiao Compound preparations. Methods: The optimum extraction conditions of Yinqiao powder were screened by orthogonal experiment. Yinqiao powder was extracted by refluxing with water, 30%, 50%, 70% and 95% ethanol respectively. The fingerprint of Yinqiao Powder was determined by HPLC. Forsythin, chlorogenic acid, arctiin, hesperidin and other substances as a process evaluation index, screening and optimization of Yinqiao powder extraction process. Results: The amount of extraction solvent and the number of extraction were the main factors that affected the extract volume of Yinqiaoqiao, followed by the extraction time. The HPLC-MS method was used to determine the content of 27 peak compounds in different extracts of Yinqiaoqiao. Conclusion: The method is simple, reproducible, and has good specificity for the identification of Yinqiaosan extract. The chromatographic peaks of Yinqiaosan are well separated and almost separated from the baseline. The Yinqiaosan Extract some of the chemical composition and content differences, as Yinqiaosan antiviral spectrum-based research.
