目的观察气候因素对Ⅱ型胶原蛋白所致类风湿关节炎(RA)模型大鼠关节炎症、免疫脏器指数及组织形态的影响。方法将40只SD大鼠随机分为空白组、Ⅱ型胶原组、Ⅱ型胶原+人工风寒湿组(简称风寒湿组)、Ⅱ型胶原+人工风湿热组(简称风湿热组)4组,每组10只,雌雄各半。空白组给予等量生理盐水,Ⅱ型胶原组皮下注射Ⅱ型胶原佐剂混合液,其余2组采用Ⅱ型胶原佐剂混合液诱导,外加人工气候因素制备RA大鼠模型。造模28 d后,比较各组大鼠的关节炎评分、胸腺指数、脾脏指数,并观察各组大鼠左后肢踝关节的组织形态改变。结果与空白组比较,各模型组大鼠均出现了不同程度的足趾关节肿胀,关节炎评分均显著升高,差异有高度统计意义(P<0.01);各模型组脾脏指数均有所升高,其中风寒湿组与空白组比较差异有统计意义(P<0.05);组织形态观察,造模后,Ⅱ型胶原组、风寒湿组和风湿热组大鼠踝关节出现不同程度的滑膜增生、淋巴细胞浸润。结论气候因素对Ⅱ型胶原蛋白所致RA模型大鼠关节炎症、组织形态具有一定的影响,能加重RA大鼠关节的病理损伤。
Objective To observe the effects of climatic factors on the joint inflammation, immune organ index and histomorphology in rheumatoid arthritis (RA) model rats induced by type Ⅱ collagen. Methods Forty Sprague-Dawley rats were randomly divided into 4 groups: blank control group, type Ⅱ collagen group, type Ⅱ collagen + artificial cold and dampness group, type Ⅱ collagen + artificial rheumatic fever group (referred to as rheumatic fever group) Each group of 10, male and female half. The rats in the blank group were given the same amount of normal saline. The type Ⅱ collagen group was subcutaneously injected with the type Ⅱ collagen adjuvant. The other two groups were induced by the type Ⅱ collagen adjuvant. The artificial climatic factors were used to prepare the RA rat model. After 28 days, arthritis score, thymus index and spleen index of rats in each group were compared, and histological changes of ankle joint in the left hind limb were observed. Results Compared with the blank group, the swelling of the toe joint and the arthritis scores of the rats in each model group were significantly increased (P <0.01), and the spleen index of each model group increased (P <0.05). After the model was established, synovial membrane appeared in the ankle joint of type Ⅱ collagen group, wind-cold-dampness group and rheumatic fever group in varying degrees Hyperplasia, lymphocyte infiltration. Conclusion Climatic factors have some effects on joint inflammation and histomorphology of RA model rats induced by type Ⅱ collagen, which may aggravate the pathological injury of RA rats.
