合成了2种三齿多吡啶钴(Ⅱ)配合物,用元素分析、IR等对它们进行了表征,用循环伏安法、电子吸收光谱、荧光光谱等方法研究了配合物与CTDNA的相互作用以及配合物对pBR322DNA的断裂作用。结果表明,配合物[Co(PhTPY)2]2+与DNA的作用属中等强度的部分插入,配合物[Co(H2Bzimpy)2]2+与DNA的作用属静电作用。凝胶电泳实验表明,用310nm光辐射15min,配合物[Co(PhTPY)2]2+可选择性地断裂pBR322DNA为开环缺口性和线型DNA,配合物[Co(H2Bzimpy)2]2+对DNA的断裂没有选择性。
Two kinds of tridentate polypyridyl cobalt (Ⅱ) complexes were synthesized and characterized by elemental analysis and IR. The interaction between the complexes and CTDNA was studied by cyclic voltammetry, electron absorption spectroscopy and fluorescence spectroscopy As well as the cleavage of pBR322 DNA by the complex. The results showed that the interaction between [Co (PhTPY) 2] 2+ and DNA was a moderate insertion, and the interaction between [Co (H2Bzimpy) 2] 2+ and DNA was electrostatic. The results of gel electrophoresis showed that the complex [Co (PhTPY) 2] 2+ could selectively break pBR322 DNA into open-notch and linear DNA with 310 nm light irradiation for 15 min. The complex [Co (H2Bzimpy) 2] 2+ Not selective for DNA fragmentation.
