目的:探讨螺旋藻激酶(SPK)对肾上腺素损伤人脐静脉内皮细胞(HUVEC)分泌人血管性假血友病因子(vWF)的影响,推测SPK抗血栓作用机制。方法:用不同浓度SPK对HUVEC预孵育后进行肾上腺素损伤,取细胞培养上清液,用酶联免疫吸附法检测vWF的含量。结果:与空白对照组相比,肾上腺素刺激的HUVEC分泌vWF明显增加,与肾上腺素模型组相比,SPK可剂量依赖性抑制损伤细胞分泌vWF,并随时间延长而逐渐明显。结论:SPK可以减少肾上腺素刺激下HUVEC分泌vWF,增强抗血栓作用。
Objective: To investigate the effect of spirulina kinase (SPK) on the secretion of vWF from adrenaline-injured human umbilical vein endothelial cells (HUVEC), and to speculate the anti-thrombotic mechanism of SPK. Methods: Adrenaline was pretreated with different concentrations of SPK and the supernatants were harvested. The content of vWF was detected by enzyme - linked immunosorbent assay. Results: Compared with the blank control group, the vWF secretion of epinephrine-stimulated HUVEC increased significantly. Compared with the model group, SPK could inhibit the vWF secretion of injured cells in a dose-dependent manner, and gradually increased with time. Conclusion: SPK can reduce the secretion of vWF by HUVECs stimulated by adrenaline and enhance the antithrombotic effect.
