目的:建立芹菜素的高效液相色谱检测方法,研究芹菜素与蛋白的相互作用。方法:采用C8柱(250mm×4.6mm,5μm)进行色谱分析,梯度洗脱,检测波长337nm,流速1.0mL·min-1;同时,结合平衡透析法,研究了芹菜素与蛋白的相互作用。结果:在2min之内实现了芹菜素的快速检测,并且在37℃的水浴恒温培养4h后,芹菜素与蛋白发生相互作用,最大结合量为5.55,结合常数为0.17(mg·L-1)-1。结论:本法可以快速准确地检测芹菜素,并可研究其与蛋白的相互作用。
Objective: To establish a HPLC method for the determination of apigenin and to study the interaction between apigenin and protein. Methods: C8 column (250 mm × 4.6 mm, 5 μm) was used for the chromatographic analysis and gradient elution. The detection wavelength was 337 nm and the flow rate was 1.0 mL · min-1. Meanwhile, the interaction between apigenin and protein was also studied by equilibrium dialysis. Results: The rapid detection of apigenin was achieved within 2 min. The apigenin interacted with the protein at a constant temperature of 37 ℃ for 4 h. The maximum binding capacity was 5.55 and the binding constant was 0.17 (mg · L -1) -1. Conclusion: This method can detect apigenin quickly and accurately and study its interaction with protein.
