目的研究小檗碱(Ber)对血管紧张素Ⅱ(AngⅡ)诱导的心肌成纤维细胞(CFB)增殖和胶原合成的影响,并探讨其作用机制。方法采用酶消化法和差速贴壁法获得纯化的CFB,应用MTT法测定CFB增殖、羟脯氨酸法测定胶原蛋白含量、化学比色法测定NO含量和NOS活力、ELISA法测定TGF-β1含量。结果小檗碱在1.25~10 mg/L浓度范围内可明显抑制AngⅡ诱导的CFB增殖和胶原蛋白合成增加,同时可升高NO含量和NOS活力、降低TGF-β1含量。结论小檗碱可抑制AngⅡ诱导的心肌成纤维细胞增殖和胶原蛋白合成的作用,对抑制心肌纤维化,改善心室重构有积极意义,其作用机制与促进心肌成纤维细胞分泌NO、减少TGF-β1含量有关。
Objective To investigate the effect of berberine on the proliferation and collagen synthesis of angiotensin II (Ang II)-induced cardiac fibroblasts (CFB) and to explore its mechanism of action. Methods Purified CFB was obtained by enzymatic digestion and differential adherent method. CFB proliferation was measured by MTT assay, collagen content was determined by hydroxyproline method, NO content and NOS activity were determined by chemical colorimetry, and TGF-β1 was measured by ELISA. content. Results Berberine significantly inhibited the proliferation and collagen synthesis of CFB induced by AngII in the concentration range of 1.25~10 mg/L. At the same time, berberine increased the content of NO and NOS and decreased the content of TGF-β1. Conclusion Berberine can inhibit the proliferation of Ang II-induced fibroblast proliferation and collagen synthesis. It has positive significance in inhibiting myocardial fibrosis and improving ventricular remodeling.
