目的 探讨因患缺氧缺血性脑病 (hypoxicischemicencephalopath ,HIE)死亡新生儿的脑组织是否存在细胞凋亡情况。方法 收集因HIE死亡新生儿 5例 (HIE组 )及非因HIE死亡新生儿 2例 (非HIE组 ) ,以肺活检穿刺针自前囟穿刺取脑组织 ,通过HE染色、原位末端脱氧核苷酸转移酶标记法 (TUNEL)及电镜等方法 ,观察脑细胞病理形态变化 ,计数单位面积下凋亡阳性细胞数。结果 HIE组新生儿脑细胞在光镜及电镜下观察可见较多凋亡细胞及坏死细胞 ,TUNEL染色凋亡细胞阳性率平均约 10 90个 /mm2 ,非HIE组新生儿脑细胞少见凋亡 ,阳性率约 3 5 0个 /mm2 ,电镜下细胞结构保存完整。结论 患HIE死亡新生儿脑细胞存在凋亡 ,从在体研究方面证实细胞凋亡参与新生儿HIE发病机制。
Objective To investigate the existence of apoptosis in brain tissue of newborn infants who died of hypoxia ischemic encephalopathy (HIE). Methods Five neonates (HIE group) died of neonatal HIE and two neonates (non-HIE group) died of neonatal HIE were collected. The brain tissue was collected from the antero-skull with a lung biopsy needle and stained with HE. TUNEL and electron microscopy were used to observe the pathological changes of brain cells. The number of apoptotic cells per unit area was counted. Results The number of apoptotic cells and necrotic cells in neonatal brain cells of HIE group was higher than that of necrotic cells. The positive rate of TUNEL-stained apoptotic cells was about 10 90 / mm2. The apoptosis of neonatal brain cells in non-HIE group was rare, Positive rate of about 350 / mm2, cell structure preserved under electron microscopy. Conclusions There is apoptosis in brain cells of newborn infants with HIE death. From the in vivo studies, it is confirmed that apoptosis is involved in the pathogenesis of neonatal HIE.
