目的 :研究血管内皮生长因子 (VEGF)和明胶酶 -A(matrixmetalloproteinase -2 ,MMP -2 )的表达与口腔鳞癌转移的相关性及VEGF和MMP -2的关系。方法 :应用Boyden小室体外侵袭实验比较口腔鳞癌细胞系TSCCa和转移癌细胞系GNM的体外侵袭力 ;采用半定量逆转录聚合酶链反应 (RT -PCR)检测两细胞系中VEGF和MMP -2mRNA表达情况 ,同时用酶联免疫吸附实验 (ELISA)的方法定量测定口腔鳞癌TSCCa细胞系和颈淋巴转移癌GNM细胞系中VEGF和MMP -2的活性。结果 :Boyden小室体外侵袭实验表明转移癌细胞系GNM的体外侵袭力较强 ,其穿膜细胞相对百分率为 (40± 3.6 ) %,而口腔鳞癌细胞系TSCCa体外侵袭力相对较弱 ,其穿膜细胞相对百分率为 (16± 3.2 ) %,两者差异具有显著性 (P <0 .0 5 ) ;RT -PCR结果显示GNM细胞VEGF和MMP-2mRNA表达水平均较TSCCa细胞高 (P <0 .0 5 ) ;ELISA法显示颈淋巴转移癌GNM细胞系中VEGF和MMP-2的活性显著高于口腔鳞癌细胞系TSCCa(P <0 .0 5 )。结论 :口腔鳞癌细胞体外侵袭力与VEGF和MMP -2的表达正相关 ;在口腔鳞癌侵袭转移中 ,VEGF与MMP -2可能具有某种内在联系 ,有可能作为反映口腔鳞癌侵袭转移的生物学指标。
Objective: To investigate the correlation between the expression of vascular endothelial growth factor (VEGF) and matrixmetalloproteinase-2 (MMP-2) and the metastasis of oral squamous cell carcinoma and the relationship between VEGF and MMP-2. Methods: Boyden chamber invasion assay in vitro was used to compare the invasiveness of oral squamous cell carcinoma cell line TSCCa and metastatic cancer cell line GNM. Semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) was used to detect the expression of VEGF and MMP-2 mRNA in both cell lines Meanwhile, the activity of VEGF and MMP-2 in oral squamous cell carcinoma TSCCa cell line and cervical lymph node metastasis cell line GNM were quantitatively determined by enzyme linked immunosorbent assay (ELISA). Results: In vitro invasion assay of Boyden chamber showed that the metastatic cancer cell line GNM had strong invasiveness in vitro and the relative percentage of penetrating cells was (40 ± 3.6)%. However, invasiveness of oral squamous cell carcinoma cell line TSCCa was relatively weak. The relative percentage of membrane cells was (16 ± 3.2)%, the difference was significant (P <0.05). The expression of VEGF and MMP-2 mRNA in GNM cells was higher than that of TSCCa cells by RT-PCR .0 5). The activity of VEGF and MMP-2 in GNM cell line of cervical lymph node metastasis was significantly higher than that of oral squamous cell carcinoma cell line TSCCa (P <0.05) by ELISA. CONCLUSION: The invasiveness of oral squamous cell carcinoma in vitro is positively correlated with the expression of VEGF and MMP-2. In the invasion and metastasis of oral squamous cell carcinoma, VEGF and MMP-2 may have some intrinsic relationship, which may reflect the invasion and metastasis of oral squamous cell carcinoma Biological indicators.
