采用新型分子标记TRAP对不同产地的丹参进行研究,建立丹参鉴别的新方法,并探索其遗传变异规律。应用CTAB法提取DNA,采用PopGene32软件进行聚类分析,并构建遗传系统树。筛选的6对引物扩增出203个条带,其中多态性条带为43,占21.1%。聚类分析结果显示4种丹参被分为2类群3亚群,其遗传一致性平均为0.0788,遗传距离平均为0.9245。实验结果表明:TRAP分子标记技术可以在分子水平上鉴别丹参的不同品种,四地丹参具有较高的遗传稳定性。
A new molecular marker, TRAP, was used to study Salvia miltiorrhiza in different habitats and to establish a new method for identifying Salvia miltiorrhiza. The genetic variation pattern was also explored. The DNA was extracted by CTAB method and PopGene32 software was used for cluster analysis. The genetic tree was constructed. A total of 203 bands were amplified by 6 pairs of primers, of which 43 bands were polymorphic (21.1%). Cluster analysis showed that four kinds of Salvia miltiorrhiza were divided into two groups and three subgroups. The average genetic identity was 0.0788 and the average genetic distance was 0.9245. The experimental results showed that: TRAP molecular marker technology can identify different varieties of Salvia miltiorrhiza at the molecular level, and the genetic stability of Salvia miltiorrhiza in four places is high.
