目的初步探讨植物甾醇诱导人神经母细胞瘤SH-SY5Y细胞凋亡的可能作用机制。方法用不同浓度(0,0.5,2.0,8.0μmol/L)的植物甾醇与SH-SY5Y细胞共同培养,流式细胞术测定细胞周期,Western-blot检测凋亡相关蛋白Caspase-9、Caspase-3的前体及活化体(Pro-Caspase-9、Cleaved-Caspase-9、Pro-Caspase-3、CleavedCaspase-3)的表达。结果不同浓度的植物甾醇可引起SH-SY5Y细胞不同程度的凋亡,并将细胞周期阻滞在G0/G1期,同时引起Caspase相关蛋白的活化。当植物甾醇浓度为2.0μmol/L时,与空白对照组相比,G0/G1期细胞所占百分比从84.35%上升到88.14%,有显著统计学差异(F=25.790,P<0.001);Caspase-3,9有活性剪切体明显增多,与空白对照组相比具有统计学差异(F=30.341,P<0.001,F=18.184,P=0.001)。结论植物甾醇可引起SHSY5Y细胞的凋亡,其机制可能与Caspase-9、Caspase-3的激活有关。
Objective To investigate the possible mechanism of phytosterol-induced apoptosis in human neuroblastoma SH-SY5Y cells. Methods Phytosterols of different concentrations (0,0.5,2.0 and 8.0 μmol / L) were co-cultured with SH-SY5Y cells. The cell cycle was determined by flow cytometry. The apoptosis-related proteins Caspase-9 and Caspase-3 (Pro-Caspase-9, Cleaved-Caspase-9, Pro-Caspase-3, Cleaved Caspase-3) Results Phytosterols at different concentrations induced different degrees of apoptosis in SH-SY5Y cells and arrested the cell cycle in G0 / G1 phase and activated Caspase-related proteins. When phytosterol concentration was 2.0 μmol / L, the percentage of G0 / G1 phase cells increased from 84.35% to 88.14% compared with the blank control group (F = 25.790, P <0.001) -3,9 active shear was significantly increased compared with the control group was statistically significant (F = 30.341, P <0.001, F = 18.184, P = 0.001). Conclusion Phytosterol can induce the apoptosis of SHSY5Y cells, which may be related to the activation of Caspase-9 and Caspase-3.
