建立了超高效液相色谱-串联质谱法测定松花粉中赤霉素的含量。松花粉提取物经80%乙醇提取、正丁醇萃取后,采用BEHC_(18)色谱柱分离,乙腈和水作为流动相进行梯度洗脱。结果表明,松花粉中赤霉素含量为0.075mg/g,在50 g/mL~250 g/mL的范围内线性关系良好(r=0.9994,n=3),平均回收率为92.2%(RSD%=1.5,n=3)。本方法操作简便、灵敏度高、稳定性好,为松花粉中赤霉素的含量测定提供了一种新的色谱技术方法。
A method for determination of gibberellic acid in pine pollen by ultra performance liquid chromatography-tandem mass spectrometry was established. Pine pollen extract was extracted with 80% ethanol, extracted with n-butanol, separated by BEHC_ (18) column and eluted with acetonitrile and water as mobile phase. The results showed that the gibberellin content of pine pollen was 0.075mg / g and the linearity was good (r = 0.9994, n = 3) in the range of 50 g / mL ~ 250 g / mL with the average recovery of 92.2% % = 1.5, n = 3). The method is simple in operation, high in sensitivity and good in stability, and provides a new chromatographic method for the determination of gibberellin in pine pollen.
